The palace was first built by the Knights of St John of Jerusalem on the side of the Byzantine Fortress of the 7th century. It was completely deserted during the Turkish occupation and in 1856 and it was completely destroyed by an explosion.

The Knight's Street used to be the official road connection the religious with the civilian centre of the Castle,which means the Latin's Cathedral with the Grand Master's Palace
The medieval architecture of Rodos is product of two eras.
The first starts from 1309 when Rodos was conquered by the Knights and lasts till 1480 when the Ottomans first besieged the city, which means more than 107 years.However, very few buildings remain from that paerio.
The second lasts from 1481 till 1522, only 42 years. During that period, the island is under the Ottoman's occupation. Most of the buildings of the old down date back to that period.

The restauration of the palace with many alterations was carried out between 1937 and 1940 under the supervision of the Iralian architect Vittorio Mesturino and the building was adorned with murals.

From the Square of the Jewish Martyrs starts the the touristic street of the Ancient Town, Aristotelous Street which leads to meeting point the Ippokratous Square. The most important building here is Castellania, the Penal Court of the Knights
