I was coming from Santa Cruz ? Bolivia. Flew Santa Cruz-Caracas, and then connected to Isla Margarita. There are charter flights from Canada and the USA; however, most flights connect in Caracas (which is a very happening place).
If you would like to take a boat you can do so from Puerto La Cruz on the Venezuelan Caribean Coast (east of Caracas, you can get to Puerto La Cruz by airplane or driving).
Within the island there are plenty of buses that will take you everywhere (very cheap), you can also take taxis but these are trickier especially if you are not Venezuelan. You can get car rental everywhere, the tourist infrastructure of the island is very good and the people are very nice.
If you want to bike, remember that the island is hilly.
