Any way you can think of! If you're flying in, there's JFK and LaGuardia Airports. If you're driving in, know where you're going before hand. The last thing you want is to be lost in NYC or be stuck in traffic trying to figure out where you're going or where you are! There are thousands of roads and thousands of expressways criss-crossing the city!
Subway is THE best way. Well, I mean, walking of course, but to go long distances, take the subway. It's very efficient, quick, and cheap, especially if you buy a visitor's day pass.
