By bus: Two Melbourne coach stations right in the city centre, one on Franklin Street, the other one on Spencer Street.
By train: Trains to and from all major cities of Australia.
Trams! The tram is the easiest way to get around if you plan to do some sightseeing around the city...remember to always carry a validated ticket, otherwise u can get a AUD$100 fine if caught by a ticket inspector. Remember to take the City Circle Tram, that one is free and specially there for tourists. It goes around the city and has good commentaries about some interesting spots on the city. If you are in a rush don't depend on the City Circle Tram for transportation, you can easily walk around the city (which is free too) or take any other tram.
Trains: Better if u plan to go to nearby cities or travel farther distances from Melbourne e.g. Werribee, Geelong, Brighton
Taxi: the most expensive way to get around.
Bus: routes are quite confusing but still fine if u can't catch the tram or train
