The airport is the most common way, but buses from Malaysia and further north are cheap, easy, and comfortable. I recommend paying the little extra to go all the way to Singapore rather than stopping in Johor Bahru and crossing on your own. It's just a hassle, and you'll just end up on a more crowded bus to save a dollar or two.
Buy a Transitlink Guide from a bookstore or the sales counters in MRT stations. Armed with this little S$1.50 book and a good map, you can get anywhere in Singapore.

Also, get a normal farecard (you can get a refund for the unused value). Forget the tourist card. It's a ripoff.
