The easist way back and forth between Hoboken and Manhattan is by train (the PATH - $1.50 per way - with one line going between 33rd street right near Penn Station, and stopping intermittantly down the western side of Manhattan - and another line between Hoboken and the World Trade Center, which also goes along to Newark).
There is also a ferry (NY Waterway - $2 per way) that leaves from the western edge of the World Financial Center and goes to the Hoboken PATH station. The ferry is much nicer than the PATH, and generally takes less than 10 minutes.
NY cabbies may or may not take you to NJ, depending on the time and the driver. If you opt to try to get a cab, keep in mind you are crossing a state line and it won't be cheap - but negotiate a price anyway (you shouldn't pay more than $25).
