1. The overnight bus for Inle departs in the early afternoon and arrives at Kalaw between 4-6AM and continues on the Shweyaung, the junction town where you get dropped off for a songthaew or shared ride to Nyaungshwe/Inle Lake town, dropping off passengers between 6AM and 8AM. It's a grueling ride but a great way to meet and interact with the Burmese.

2. No problem in visiting the destinations you've listed with the EXCEPTION of Ngapali in 2 weeks. That would be 'pushing' it, especially if you encounter bus mechanical problems One of the best airfare deals/routes is flying from Heho near Inle to Thandwe for Ngapali and then flying back to Yangon from there.

Thus, you could reverse your route, going to Pyay>Bagan/Nyuang U>Mandalay>Inle and fly to Ngapali and back to Yangon in 15 days.
