OK, as far as transportation goes, it's usually time versus money. If you are really on a budget but have lots of time then take the bus. Several options here, anywhere from taking local forms of busses which could take you up to two days, BKK to Phuket, several transfers and sore backsides. Not recommeded! Or the delux busses which will still take you a long time, more comfortable but how comfortable can you be on a 10-12 hour bus ride.
You could take the train from BKK to Surat Thani, then a bus to Krabi or Phuket. The overnight sleeper on the train will save you hotel costs and it's supposed to be quite comfortable, but you'll still end up on a bus for several hours to your destination.
Air is the fastest and it's really not that expensive. Direct flights from BKK right into Krabi or Phuket, You can normally get air for two, O/W for about 150$US, 75 each. www.airasia.com

If you are planning to go to Singapore first, then I would either take a flight into the south from Singapore, go to Phuket/Krabi, then hit Bangkok after, save you time and it's a lot easier. You can also take a train from Singapore to southern Thailand.

As far as where to go. I was not impressed with Krabi, mostly because of the beaches. However you can get to some of the nearby islands that have great beaches. I liked Phuket better. It really all depends on what YOU are looking for in your travels as to where you decide to go. Are you looking for some places to party? nice quiet get away? etc... We loved Khao Lak. Nice and quiet. The beaches were great and the town is still pretty quiet and you can find some great accomodations. Let me know if that interests you and I can direct you to a great place in Khao Lak. The water on this side versus Krabi is (in my opinion) far better, and after all, you are going to the south for the beaches, right? All of the islands are still accessible from Phuket and Khao Lak.
Good luck deciding and have a great trip! let me know if you need anything else.
