Though there is a Heliport on the island and some tour operators will do a helicopter transfer from Malta to Gozo, most people arrive on the island by ferry from Malta. The ferries run every 45 minutes throughout the day, less regularly at night - but still they run - from Mgarr on Gozo to Cirkewwa on Malta. Cirkewwa is the closes point on mainland Malta to Gozo.
Fares are reasonable (for foot passengers - no idea about cars) and the crossing takes about 25 minutes.
There is a ferry directly from Valletta (well, Sa Mainson, near Valletta) too, but this only runs about once per day and you have to "ring for details".
