If you're committed to your car, then you can of course drive to Stolpen. For the countryside around the town including the cute village Lauterbach e.g. the car is a good option anyway.
Stolpen is only 5 km from the major road B6 Dresden - Bautzen - Görlitz. From Dresden's outskirts you drive about 20 minutes. From Pirna the road is rather windy and narrow, driving time is about 20 minutes also.
The issue I have with driving to Stolpen (ecological problems aside) is parking. There are only VERY few free parkings, one at the side of the one-side street right after entering the town (but still outside the old town) from Dresden. That one has a time limit (don't forget the parking disk!) of one hour. The other is the parking lot of the Netto grocery store on the eastern edge of the town (direction Neustadt) - but it is a bit risky as the parking is originally meant (and marked with signs) for shoppers only. I haven't heard of someone getting fined or even towed, though.
The other parkings are signposted but NOT FREE. Not overly expensive, though, but still ... Most central is the parking (not many lots, though) on the historic market square. The problem here is that in winter you might have a problem with snow and ice ... it's sloping. See pic 2!

Another option to get to Stolpen by public transportation is the train. Stolpen is one the train route Pirna - Neustadt (Sachs) - Bad Schandau. The leg Pirna - Stolpen is not overly exciting (countryside, rather flat) but especially beyond Neustadt and closer to Bad Schandau the trip is quite scenic with views of the National Park Saxon Switzerland, through a lovely valley and passing numerous bridges + seven tunnels. Kids love that :-)
The trains Pirna - Stolpen - Neustadt run hourly till late in the evening (weekdays, on weekends only every other hour, like between Neustadt and Bad Schandau all week), in Pirna you have timed connections to the commuter trains along the Elbe river valley to Dresden and Meißen.
The problem with the trains in Stolpen is that the train station is quite a distance from the old town/castle. It's about a 20 minutes walk, I'd say - and a bit uphill, too.
Fares are the same as for the bus - tariffs of the VVO network apply.

The most convenient travel mode to Stolpen is IMO the bus. Stolpen is in the public transportation network VVO, just like Dresden. The bus line 261 runs at least hourly from Monday to Friday, every two hours on weekends/holidays, the route Dresden - Stolpen - Neustadt - Sebnitz (where you can change to the buses for the National Park Saxon Switzerland). Another bus line connects Stolpen with Pirna - a little less frequently.
The villages around Stolpen which are part of the municipality are either along the bus routes mentioned above (Stolpen-Altstadt, Langenwolmsdorf, Helmsdorf) or they get very poor bus service, sadly (Lauterbach, Helmsdorf).
There are two bus stops in the town Stolpen, both are fine for a visit. "Schützenhausstraße" is the first one coming from Dresden, best if you want to explore the old town first, "Ärztehaus" is the other, perfect for the quick walk up to the castle.
The single-trip fare (one way) from Dresden to Stolpen is (in 2012) 5.60 Euro (3 zones). From Pirna you pay 3.80 Euro (2 zones).
