If you are taking the Wine Route, surely you will be visiting a number of wineries.

Unique Suggestions: If you intend to try a lot of wines, avoid swallowing it. Otherwise you may not be able to drive.
Don't start driving with an empty stomach. Of course in some wineries they might offer you some nuts or some soutzouko, but it is so small quantity that you might easily get drank if you have eaten nothing.

Fun Alternatives: Let the wine run down the route to your stomach - preferable accompanying it with some food.

Savour the taste it leaves at your your plate, what is known as "aftertaste".
